2,4-二硝基氯苯诱导 BALB / c 小鼠特应性皮炎模型的建立

Establishment of Atopic Dermatitis Model Induced by DNCB in BALB / C mice

摘要: 目的 利用 2, 4-二硝基氯苯 ( DNCB) 诱导 BALB/c 小鼠建立特应性皮炎 ( AD) 模型, 探讨最适条件。方法 给予 BALB/c 小鼠背部皮肤不同药物浓度以及不同的造模时间致敏及激发,在末次给药后,评估小鼠背部皮肤状态。 取背部皮肤组织做组织病理学检测,分离血清检测免疫球蛋白 E( IgE) 水平,制备皮肤匀浆液检测细胞因子白细胞介素-4( IL-4) 、白细胞介素-5( IL-5)及白细胞介素-13( IL-13)分泌水平。 结果 4 组造模模型组动物中,D组小鼠 0. 5% DNCB 致敏 3 次,1% DNCB 激发 6 次,其皮肤皮炎评分最高,病理结果显示皮肤结构不完整,棘层明显增厚,细胞内水肿明显,大量炎性细胞浸润,血清 IgE 水平升高,皮肤组织细胞因子明显升高,相比于其他模型组其各种检测指标更接近特应性皮炎皮肤改变。 结论 早期低剂量致敏,后期高剂量激发可成功建立小鼠 AD 模型,这为进行 AD 的相关科学研究提供了一定的实验依据。

Abstract: Objective To establish atopic dermatitis ( AD ) model in BALB / c mice induced by 2, 4- dinitrochlorobenzene (DNCB) , and to explore the optimal conditions, so as to provide theoretical basis for the establishment of animal model in the research of AD. Method BALB / C mice were sensitized and stimulated with different drug concentrations and different modeling time. After the last administration, the severity of dermatitis in the dorsal skin of DNCB-induced AD mice was evaluated based on symptoms, and hematoxylin and eosin ( H&E) and toluidine blue ( TB ) staining were carried out for further analysis. Serum samples were collected for immunoglobulin E ( IgE) detection, and the skin homogenates were prepared to detect the levels of interleukin-4 ( IL-4) , interleukin-5 ( IL-5) and interleukin-13 ( IL-13) . Result Mice in group D were sensitized by 0. 5% DNCB for 3 times and stimulated by 1% DNCB for 6 times. The skin severity score was the highest in group D. The pathological result showed that the skin structure was incomplete, the Spinous layer was obviously thickened, the intracellular edema was obvious, and a large number of inflammatory cells were infiltrated, the levels of serum IgE and cytokines in skin tissue were significantly higher than other groups. Conclusion Low-dose sensitization at early stage and high-dose stimulation at late stage can establish mouse AD model successfully, which lays a foundation for further study on AD.
